Background

Thisantibodyisdesigned,produced,andvalidatedaspartofacollaborationbetweenRocklandandtheNationalCancerInstitute(NCI)andissuitableforCancer,ImmunologyandNuclearSignalingresearch.ActivatorofHsp90ATPase(AHA1)stimulatestheinherentATPasecycleofHsp90,whichisessentialforitschaperoneactivityinvivo. Theactivationand/orstabilityofmanyofthekeyregulatoryandsignalingproteinsoftheeukaryoticcelldependontheirinteractionwiththeHsp90molecularchaperone. Hsp90isassistedandregulatedbyco-chaperonesthatparticipateinanorderedseriesbothtoassistclient-proteinrecruitmentorreleaseandtomodulateprogressthroughtheATPasecoupledchaperonecycle. StructuralanalysisandmutagenesisshowthatbindingoftheN-terminaldomainofAHA1toHsp90promotesaconformationalswitchinthemiddle-segmentcatalyticloop(aa370–390)ofHsp90thatexposesthecatalyticArg380andenablesitsinteractionwithATPintheN-terminalnucleotide-bindingdomainofthechaperone. RecentstudiesshowthatAHA1modulatesHsp90-dependentstabilityofthefoldingofthecysticfibrosistransmembraneconductanceregulator(CFTR)intheendoplasmicreticulum(ER). Down-regulationofAHA1rescuesmisfoldingofCFTRincysticfibrosis.
