| Description | Alphasynucleinisanabundant140aminoacidneuronalprotein,expressedprimarilyatpresynapticterminalsinthecentralnervoussystem.Alphasynucleinhasbeenassociatedwithseveralneurodegenerativediseases.Apointmutationinthegenecodingforthealpha-synucleinproteinwasthefirstdiscoverylinkingthisproteintoararefamilialformofParkinsonsdisease(PD).Subsequently,othermutationsinthealpha-synucleingenehavebeenidentifiedinfamilialPD.TheaggregatedproteinaceousinclusionscalledLewybodiesfoundinPDandcorticalLewybodydementia(LBD)werediscoveredtobepredominantlyalpha-synuclein.Aberrantaggregationofalpha-synucleinhasbeendetectedinanincreasingnumberofneurodegenerativediseases,collectivelyknownassynucleopathies.Alpha-synucleinexistsphysiologicallyinbothsolubleandmembrane-boundstates,inunstructuredandalpha-helicalconformations,respectively.Thephysiologicalfunctionofalpha-synucleinappearstorequireitstranslocationbetweenthesesubcellularcompartmentsandinterconversionbetweenthe2conformations.Abnormalprocessingofalpha-synucleinispredictedtoleadtopathologicalchangesinitsbindingpropertiesandfunction. |
