PRAS40(proline-richAkt/PKBsubstrateof40kDa)actsattheintersectionoftheAkt-andmTOR-mediatedsignalingpathwaysanditsphosphorylationbyAktandbymTORC1resultsindissociationofPRAS40frommTORC1whichmayrelieveaninhibitoryconstraintonmTORC1activity(Wizaetal.,2012).PhosphorylationofPRAS40byAktandassociationwith14-3-3hasalsobeenindicatedtobecrucialforinsulintostimulatemTOR.(VanderHaaretal.,2007).TheprimaryfunctionofPRAS40invivoinDrosophilahasbeenshowntoregulateTORC1activity,andnottoactasadownstreamtargetandeffectorofTORC1(Pallaresetal.,2012).
